تم تحضير وتقييم سلسلة جديدة من الهجائن غير المتماثلة من الكومارين–1,3,4-ثياديازول (T1a–T1g، T2a–T2c وT3a–T3b) بيولوجيًا بهدف استهداف بعض المسارات الكيميائية الحيوية المرتبطة بمرض الزهايمر. حُضّرت هذه المركبات انطلاقًا من ثلاثة طليعاتللكومارين (C1–C3)، تلتها خطوة ألكلة للحصول على وسائط كومارينيةمؤلكلة، ثم تكثيفها مع الثيوسيميكاربازيد تحت ظروف قاعدية معتدلة مما أدى إلى تكوين هجائن جديدة غير متماثلة. تم الحصول على اثني عشر مركبًا بمردود مرتفع (>82%)، وتم توصيفها باستخدام مطيافية الرنين المغناطيسي النووي RMN ¹Hو ¹³C. أظهرت التقييمات البيولوجية نشاطًا مضادًا للأكسدة ملحوظًا بالإضافة إلى فعالية مثبطة لإنزيم أستيل كولين إستراز (AChE). وقد أظهرت المركباتT1a–T1d وT1f وT2b قدرة عالية على اصطياد الجذور الحرة (IC₅₀ = 24.32–50.14 µM)، في حين أظهر اختبار الاختزال باستعمال o-فينانثرولين أن المركبات T1f وT3a وT1d هي الأقوى اختزالًا (A₀.₅ = 11.38–31.90 µM). أما فيما يخص تثبيط إنزيم AChE، فقد سجل المركبان T1g وT1f أفضل النتائج (IC₅₀ = 20.35–38.90 µM). كما أكدت الدراسات الحاسوبية، بما في ذلك الالتحام الجزيئي وحسابات الكيمياء الكمية (DFT) وتنبؤات ADMET، وجود تفاعلات قوية مع الموقع الفعال للإنزيم وخصائص إلكترونية ودوائية حركية ملائمة. ومن بين المركبات المدروسة، برزت T1c وT1f وT1g وT3a بفضل الجمع بين نشاطها المضاد للأكسدة وقدرتها على تثبيط إنزيم AChE، مما يجعلها مرشحات واعدة لتطوير عوامل علاجية محتملة ضد مرض الزهايمر والأمراض التنكسية العصبية الأخرى المرتبطة بالإجهاد التأكسدي.
De nouveaux hybrides asymétriques coumarine–1,3,4-thiadiazole (T1a–T1g, T2a–T2c et T3a–T3b) ont été synthétisés et évalués biologiquement dans le but de cibler certaines voies biochimiques impliquées dans la maladie d’Alzheimer. Les composés ont été préparés à partir de trois précurseurs de coumarine (C1–C3), suivis d’une étape d’alkylation permettant d’obtenir des intermédiaires coumariniquesalkylés, puis d’une condensation avec la thiosemicarbazide sous des conditions basiques modérées conduisant à la formation de nouveaux hybrides asymétriques. Douze composés ont été obtenus avec d’excellents rendements (>82 %) et caractérisés par spectroscopie de résonance magnétique nucléaire RMN ¹H et ¹³C. Les évaluations biologiques ont révélé une activité antioxydante significative ainsi qu’une activité inhibitrice de l’acétylcholinestérase (AChE). Les composés T1a–T1d, T1f et T2b ont présenté une forte activité de piégeage des radicaux libres (IC₅₀ = 24,32–50,14 µM), tandis que le test de réduction à l’o-phénanthroline a identifié les composés T1f, T3a et T1d comme les meilleurs agents réducteurs (A₀.₅ = 11,38–31,90 µM). En ce qui concerne l’inhibition de l’AChE, les composés T1g et T1f ont montré les meilleures activités (IC₅₀ = 20,35–38,90 µM). Les études in silico, comprenant le docking moléculaire, les calculs de chimie quantique (DFT) et les prédictions ADMET, ont confirmé de fortes interactions avec le site actif de l’enzyme ainsi que des propriétés électroniques et pharmacocinétiques favorables. Parmi les composés étudiés, T1c, T1f, T1g et T3a se sont distingués par la combinaison de leurs activités antioxydantes et inhibitrices de l’AChE, faisant d’eux des candidats prometteurs pour le développement de nouveaux agents thérapeutiques potentiels contre la maladie d’Alzheimer et d’autres maladies neurodégénératives associées au stress oxydatif.

Compounds T1a–T1d, T1f, and T2b exhibitedstrong free-radical scavengingactivity (IC₅₀ = 24.32–50.14 µM), while the o-phenanthrolinereducing power assayidentified T1f, T3a, and T1d as the mostpotentreducing agents (A₀.₅ = 11.38–31.90 µM). RegardingAChE inhibition, compounds T1g and T1f displayed the highestactivities (IC₅₀ = 20.35–38.90 µM). In silico studies, includingmoleculardocking, densityfunctionaltheory (DFT) calculations, and ADMET predictions, confirmedstrong interactions with the enzyme active site togetherwith favorable electronic and pharmacokineticproperties. Among the investigated compounds, T1c, T1f, T1g, and T3a emerged as the mostpromising candidates owing to theircombinedantioxidant and AChEinhibitoryactivities, highlightingtheirpotential as lead compounds for the development of noveltherapeutic agents againstAlzheimer'sdisease and otherneurodegenerativedisordersassociatedwithoxidative stress. |
